چکیده

Three commerciallyavailablekitsfordetermining serum thyroxine (T4) by column were evaluated. Results with all were less than satisfactory, but a series of changes in the Oxford method resulted in increased reproducibility, substantially improved recovery, and extended linearity of the standard curve. Results compared well with the thyrometabolic status of patients and normal individuals. The 95% confidence limits for 219 “normal” hospital employees were 2.9-6.2 g/100 m. The suggested normal range for 241 clinically euthyroid patients is 2.5-6.0 g/100 ml. The coefficient of correlation between results of the proposed method and of the PBI method was 0.30; for the T4 by the Murphy-Pattee method it was 0.75.

Evaluation of new kits for the assessment in vitro of thyroid function by determination of serum total thyroxine, free TBG capacity, and free thyroxine index using Sephadex G-25 and '25I-labelled triiodothyronine and thyroxine

SYNOPSIS A trial was carried out on 134 patients of new kits (Ames Co) using columns of Sephadex G-25 for the determination of serum total thyroxine (Tetralute test) and for the indirect estimation of serum free thyroxine-binding globulin capacity (Trilute test). Both new methods were quicker and easier than the reference resin methods and of similar precision. The two measurements when combine...

Enzyme immunoassay for thyroxine and triiodothyronine in human serum, with use of a covalent chromatographic separation method.

A practicable enzyme immunoassay for measurement of thyroxine and triiodothyronine in serum was developed, involving a separation method based on the thiol-disulfide interchange reaction. Serum samples at alkaline pH were incubated for 1 h with antibodies and antigens labeled with beta-D-galactosidase. Each reaction mixture was then passed through a 0.1-mL column containing (anti-IgG) antibody ...

Evaluation of Total Antioxidant, Total Calcium, Selenium, Insulin, Free Triiodothyronine and Free Thyroxine Levels in Cows with Ketosis

Ketosis is an important metabolic disease of high milk-producing cows. There are significant changes in many metabolite and hormonal concentrations in metabolic diseases. This study was carried out to assess the concentrations calcium (Ca), selenium (Se), total antioxidant (TAOC), insulin, free triiodothyronine (fT3) and free thyroxine (fT4) in cows with subclinical and clinical ketosis. This s...

Candidate reference method for total thyroxine in human serum: use of isotope-dilution liquid chromatography-mass spectrometry with electrospray ionization.

BACKGROUND There is a need for a critically evaluated reference method for thyroxine to provide an accuracy base to which routine methods can be traceable. We describe a candidate reference method involving isotope-dilution coupled with liquid chromatography/mass spectrometry. METHODS An isotopically labeled internal standard, thyroxine-d(5), was added to serum, followed by equilibration, pro...
